Research Abstract |
1) Establishment of methods for partial hepatic resection and preservation We have established a technic of partial (20-50%) hepatic resection without hemorrhage or ischemia in rats and pigs. We have developed "Futhan rines preservation solution", with whchi a rat liver graft can be preserved for 48 hours. It preserved longer than UW solution (24 hours). This solution was like the extra-cellular fluid electrolytic composition which had low potassium level. 2) Hepatic cellular growth factor and its application to the hepatic transplantation Hepatic cellular growth factors were produced in the pig partial hepatic resection model. As their titers were somewhat unstable, we have not identified a proper growth factor in pig. 3) Heterotopic abult partial hepatic transplantation We established a model of heterotopic abult partial hepatic transplantation in pigs, in which a partial (50%) hepatic graft was planted in the infra-hepatic region of a recipient. nction in the recipient recipient occlusion of its hepatic artery and portal vein. 4) Production of anti-leukocyte adhesion molecule antibodies and their application to the partial hepatic transplantation Protection of hepatic wam ischemia : Male Wistar rats received partial (70%) hepatic ischemia for 90 min. Five minutes before the ischemia, monoclonal antibody (mAb) against ICAM-1, CD11a, or CD18 was administered intravenously. Then, these mAbs suppressed the infiltation of PMNs, reduced hepatic MDA level, increased ATP resynthesis and significantly increased the survival rate of rats. Endotoxin Shock : Lipopolysaccharide (LPS : 30 mg/kg) was administered to ICR mice intraperitoneally, and 1mg/kg mAb (anti-CD11a, anti-CD11b, anti-CD18, anti-ICAM-1 or anti-LECAM-1) or 30 mg/kg methylprednisolone (MP) was administered intravenously, simultaneously. These treatments significantly increased the survival rate of mice, and mAb(anti-CD18) and MP reduced the hepatic MDA level.
