成長因子投与による変形性関節症の関節軟骨修復の試み

| Research Abstract |
変形性関節症における関節軟骨欠損に対して成長因子を利用した修復についていくつかの実験をおこなってきた。本年度も関節内投与実験、関節液での軟骨病態マーカーについて、さらに軟骨細胞培養における成長因子の影響につき検討をすすめ、その成果を発表している。正常関節軟骨組織への成長因子投与実験については、幼弱ラット、成人マウスのそれぞれの膝関節への影響について実験を終え、現在、論文投稿中である。どちらの動物においても増殖性変化を促進することが明らかになっている。関節症動物モデルでの成長因子投与実験については、同増殖性変化を促進するのみならず、破壊的変化にも影響を与えることを示唆する結果が出ており、今後さらに検討を重ねる必要があると思われる。種々の軟骨組織における成長因子の発現につては成長軟骨での検討がおわり、いくつかの成長因子が成長軟骨で発現され、その細胞増殖屋、細胞分化の局所性制御因子として働いていることが示唆された。同実験の一部は論文として発表した。ヒト変形性股関節症での血清中軟骨病態マーカーについてはサンプルを採取している段階である。ヒト及び動物軟骨細胞培養での成長因子の影響については、関節症に関与のあるinterleukin産生への影響について検討した。ある成長因子が同産生を促進していることを示唆する結果がでており、関節症に対して影響する可能性があることが判明した。
