Research Abstract

It was examined whether the gene function of mouse osteoblastic MC3T3 - E 1 Cells (E 1 cells) was affected by a low-calcium environment.
In the E 1 cells with the treatment of FBS or EGF,c-fos mRNA expression increased with time in the control and low Ca groups, reached maximum at 30 minutes and then decreased at 60 minutes. The degree of the expression was significantly higher in low Ca group than in control group (p<0.01).
Since EGF activates tyrosine kinase of receptor type, it was speculated that a tyrosine kinase is involved in this enhancement in low Ca group.
When the E1 cells was pretreated with EGF + genistein which was one of tyrosine kinase inhibitors for 30 minutes, the enhancement in the E 1 cells with treatment of only EGF in low Ca group was inhibited, and the expression level approached to control level.
From these facts, it was suggested that c-fos mRNA expression is intimately lelated with the tyrosine kinase activity. Namely, E 1 cells coped with the low-calcium environment to restore to normal function against an abnormal one.
