Research Abstract |
Recent studies has clarified that cementum proteins are involved in periodontal tissue regeneration, especially new attachment to the exposed root surface. We have purified cell growth factor derived from a human tooth cementum (PCDGF) by gel permeation on the like molecular fractionation. PCDGF has following properties : (1) Molecular weight (by gel permeation) 75,000 (]SY.+-。[) 5,000 (2) Isoelectric point 6.5 (]SY.+-。[) 1.0 (3) Thermal stability PCDGF does not in activity by heat treatment at 60゚C for 30minutes. (4) Physiological activity PCDGF has chemoattractive potency to gingival fibroblasts. (5) The chemoattractive activity was not destroyed by antibodies to PCDGF,FN,TGF-beta, GF,VN. (6) SDS-polyacrylamide gel electrophoresis revealed that HPLC fraction eluted by 0.5M NaCl contained a protein with molecular weight 75kDa. These data indicated that PCDGF is novel proteins which play an important role in periodontal tissue regeneration.
