Research Abstract |
It has been interested that cell surface glycocongugates has drametically changed during differentiation and some of those were identified and clarified in the structural analysis in detail. This research was focused to the setablishment of the analytical method of small amount of glycocongugates at cell surface and its developmental application. In 1992, this method was mainly used to the analysis of glycolipids composition in the menbrane of nuclear and mitochondoria. The high potentiality of the method has been proved for the analysis of snall amount of glycolipid on both organera cell surface in 1993, based on the result, affinity chromatography combined with specific carbohydrate chain identified in each organera menbrane was produced and used to purify the protein which is able to specifically bind to the ligand using fetal rat brain as protein source. The purified proteins showed the molecular weight of about 50 kd on SDS-PAGE.Large scale purification is now performed, characterized in detail. To understand the physiological interaction between specific sugar chain ligand and binding protein, expression cloning system using mammalian cell were tried. The fundamental condition for the cloning was completely established using panning and FACS method. This system can abusolutely serve to the detection of the specific carbohydrate on cell surface, leading to the understanding between cell-cell recognition.
