Research Abstract	ヒト末梢血好中球は刺激に対する応答性が弱く、サイトカインなどによるpriming作用によって著しく増強される。しかし、種々の研究にもかかわらず、現在尚このpriming作用の分子機構は明らかにされていない。本研究では、TNF-αやG-CSFによるヒト末梢血好中球のprimingによるスーパーオキサイド生成の増大を中心にその解析を試み、このpriming作用が受容体を介した刺激応答性を特異的に増大し、またそのprimingの過程において好中球タンパク質チロシン残基のリン酸化が並行して増大し、それがpriming機構に何らかの関係を持つことを示唆する結果を得た。また、種々の非ステロイド系抗炎症性薬物のpriming過程に対する作用の解析から、タンパク質のチロシン残基リン酸化の阻害がpriming阻害と並行して進行し、primingにおけるタンパク質チロシンリン酸化の重要性をより明瞭した。更に、この解析において、priming過程にGTP-結合タンパク質も関与していることを示唆した。以上本研究においてはprimingにおけるtyrosine kinaseの関与を明らかにしたが、今後、特にprimingに並行してリン酸化の進む115kDaや120kDaタンパク質の性格を明らかにすることが出来れば、その反応の分子機構をより詳細に解析できると考えられる。
