軟部組織の組織球性増殖の本態と細胞動態に関する研究

Research Abstract	1.人体材料の臨床病理学的検索:すでに当教室および関連機関における生検および手術材料から軟部組織の組織球性増殖性疾患を抽出し,各種特種染色を施行して光顕的に検索した。予後追跡調査についても進行中であり,良性病変の中にも再発するものを見出した。 2.免疫組織化学的検索:各種の単球-組織球マーカーおよび間葉系細胞マーカーを用いて,前記の人体材料を免疫組織化学的に検索した。従来線維性組織球性腫瘍fibrohistiocytic tumorといわれていた病変の多くは間葉系細胞の増殖が主体であることがわかった。(Iwasaki H et al:Malignant fibrous histiocytoma:proliferative compartment and heterogeneity of “histiocytic" cells.Am J Surg Pathol 16:735-745,1992) 3.細胞倍養系の確立:ヒト軟部の組織球性増殖病変から培養細胞系を確立し,種々の分化抗原および細胞マーカーについて検討したところ,これらの細胞は組織球類似の機能をある程度有してはいるが,真の組織球ではなく,facultative histiocyteであることがわかった。(Iwasaki H et al:Malignant fibrous histiocytoma:A tumor of facultative histiocytes showing mesenchymal differentiation in cultured cell lines.Cancer 69:437-447,1992) 4.染色体分析:本疾患の新鮮手術材料から,分裂中期の細胞の核型を分析したところ,一部の例で多数のマーカー染色体を有する複雑な異常が認められた。(Cancer69:437-447,1992) 5.細胞動態の分析:新鮮手術材料およびパラフィン切片について抗PCNA/cyclinモノクローナル抗体で染色して増殖細胞の分布を調ベたところ,間葉系の増殖細胞の核は高頻度に標識されたが,反応性の正常組織球はほとんど標識されなかった。これによって本病変の本態は間葉系細胞を主体とする増殖性病変であることが明らかになった。(Am J Surg Pathol 16:735-745,1992)
