Research Abstract

The effecte of splenectomy and thymectomy on experimental autoimmune myasthenia gravis (EAMG) were investigated. EAMG was made in six-week-old female Lewis strain rats immunized with purified acetylcholine receptor (AChR) protein from torpedo carifornica. EAMG rats underwent sham operation, splenectomy, thymectomy, or both, one week after preparation.
Splenectomy group indicated lower titer of anti-AChR anti bodies in sera and in supernates of the culture of lymphocytes from lymphnodes than sham operation group. Lymphocyte proliferative responses to AChR,phytohemagglutinin (PHA), and lipopolysaccharide (LPS) were similar.
Splenectomy group indicated significantly rapid body weight gain and significantly high ability of movement than sham operation group. Thymectomy group also indicated these effects of operation but they were not significant.
Modification of lymphocytes subclass which was confused by EAMG was observed in splenectomy group. It was not found in both thymectomy group and sham operation group.
These results suggest that splenectomy on EAMG was effective and the effectiveness may occur by modification of T lymphocytes. The effect of thymectomy was not demonstrated significantly, presumably due to different onset mechanism of EAMG from human MG.
