泌尿器科領域悪性腫瘍における癌関連遺伝子群の異常に関する研究

Research Abstract

腎摘除術にて得られた腎癌材料を対象に、c-myc、c-fosおよびHarvey ras遺伝子のmRNAの発現状況を検討した。腫瘍mRNAにおいてc-myc遺伝子は23例中11例(48%)に、c-fos遺伝子は6例(26%)に、Harvey ras遺伝子は1例(4%)に正常組織mRNAと比較し3倍以上の過剰発現を呈した。Harvey ras遺伝子の過剰発現を示した症例は同時に他の二つの癌遺伝子の過剰発現を伴っていた。また、c-myc、c-fos遺伝子両者の過剰発現が5例(22%)にみられた。組織学的悪性度、細胞型および臨床病期と各遺伝子の過剰発現との明らかな関連性は認められなかったが、これらの遺伝子過剰発現は腎癌の癌化機序に関与しているものと推察された。手術的に得られた腎癌24例、前立腺癌18例、腎盂移行癌11例にRFLP分析を施行し、遺伝子の増幅、おるいは癌抑制遺伝子の不活化の有無につき検討した。c-myc、c-fos、Harvey ras、EGFRいずれの癌遺伝子も各腫瘍型でDNAコピー数の過剰増加は認められなかった。
