Research Abstract

Interacition between cell cycle regulatory genes anti-oncogenes contributed to the malignant potential of hydatidiform mole.
1) Expression of oncogenes and anti-oncogenes in hydatidiform mole, choriocarcinoma, and choriocarcinoma cell lines.
Northern blot analyzes have revealed elevated expression of several oncogenes and decreased expression of a few anti-oncogenes in hydatidiform mole, choriocarcinoma, and choriocarcinoma cell lines. DNA deletions of a few anti-oncogenes were also detected in hydatidiform mole by southern blot analyzes. In order to detect point mutaions of oncogenes and anti-oncogenes, SSCP method was performed. cDNA of those tumors was synthesezed and used for SSCP.SSCP has revealed several point mutations in those tumors. Those mutations were confirmed by sequencing method. Some of those mutations were detected in so-called hot spot. The others were not in hot spot.
2) Establishment of primaly culture system for early pregnant chorion.
Chorion was separeted in the trophoblast by collagenase prepatation. Primary culture system of separeted trophoblast on collangen coated petri-dish was established.
