Research Abstract |
A 6kb fragmant of the 5'-flanking region of the mouse EGF gene was investigated to characterize hormone response elements. A restriction map with 7 endonucleases was prepared and a series of chloramphenicol acetyltransferase reporter plasmids containing the 5'-flanking region-deletion mutants were constructed. An eukaryot expression vector of mouse androgen receptor was also constructed, and the expression of the receptor in COSI cells was confirmed by the enzyme-linked antibody-staining method with the anti-andogen receptor monoclonal antibody. We found a novel thyroid response element (EGF-TRE) at around position -250 : EGF-TRE matched the TRE of the Moloney leukemia virus gene by 67% in nucleotide sequence and competed specifically with the palindromic TRE for thyroid-receptor binding. The chloramphenicol acetyl-transferase assay also demonstrated a tyroid-dependent transcription activation within the region from position -400 to position -1. This region also responded to andorogen. These results suggest the existance of multicomponent regulatory complex in the EGF gene.
