Research Abstract

This study undertaken to evaluate the usefulness of bioabsorbable polymer (BAP) as a crrier of bone morphogenetic protein (BMP). Three kinds of granular BAPs (two polylactic acids with different molecular weight and lactic-glycolic acid copolymer) were implanted into rat tibiae and light microscopic sections were prepared following surgery. BAPs were gradually absorbed and replaced by bone marrow tissue by 24 weeks. Phagocytic reactions to the BAPs were noted, however, severe inflammation was not seen. Nextly, BAPs were mixed with demineralized bone particles prepared from rat femurs and laid subcutaneously. Histological examination revealed cartilage formation at 2 weeks and new bone formation at 4 weeks. Extensive bone and marrow formation were observed at 24 weeks. These results demonstrate that good tissue compatibility of BAP at the skeletal site and it dose not have negative influence on the osteinductive activity.
Crude BAP was extracted from the rat demineralized bone powder by 4M Guanidine HCl. BMP was mixed with BAP, then implanted in rat subcutaneously.New bone trabeculae with the BAP granulues occured at 2 weeks. Histomorphometrical analysis revealed that the amount of new bone of the BAP-BMP composite was more than of the BMP alone.
These results indicate that BAP is effective as a carrier of BMP.
