Research Abstract	本研究では循環調節に対するタキキニン・ペプチドの生理的役割に焦点を当てて一連の研究を行なった。前年度の研究において,タキキニン・ペプチドの中枢性血圧調節機構に関して明らかにすることができたので,最終年度(5年度)はタキキニン・ペプチドの体液調節機構について研究を進めた。ラット脳室内にNK-3受容体作動薬のセンクタイドを投与すると,容量に依存して抗利尿作用を発現した。この抗利尿作用は,バソプレッシンV2受容体拮抗薬(OPC-31260,10mg/kg)の静脈内投与により抑制された。また,センクタイドの脳室内投与により,血中のバソプレシン量が著しく上昇した。以上の結果から,中枢における体液調節機構にタキキニン・ペプチド-AVP系が存在関与し,センクタイドは室傍核のNK-3型受容体を刺激し,下垂体からバゾプレシンを遊離させ抗利尿作用を発現することが示唆された。本研究によりタキキニン・ペプチドが中枢から血圧・体液調節を行なっていることが明らかにされた。さらに,最近非ペプチド性受容体拮抗薬が開発されてきたので,ペプチドの生理的意義や病態での動態を考えるとき,臨床的効果も大いに期待できると思う。
