| Research Abstract |
研究計画にのっとりPlasminogen activator inhibitor-1(PAl-1)の構造と機能の関連について検討した。PAl-1のターゲットとなる分子であるtissue-type plasminogen activator(tPA)とPAl-1、urokinase-type plasminogen activator(uPA)とPAl-1の分子反応機作について検討し、tPA及びuPAの非触媒ドメインに可逆的PAl-1結合部位を見いだし、これらの部位とPAl-1の反応がPAl-1によるtPAuPAの即時的阻害反応に重要であることを報告した。またPAl-1を結合しPAl-1の組織での反応に重要であると考えられているvitronectinのPAl-1の結合部位を検討した結果、7アミノ酸残基からなるペプチドセグメントがPAl-1との結合反応に重要であることを明かにし報告した。また、PAl-1を酵素で限定分解したのち逆相HPLCでPAL-1フラグメントを分取し、PAl-1フラグメントを検討した結果、PAl-1分子内のtPAのfinger domain結合部位を明かにしえた。
