| Research Abstract |
アミノ酸ラセマーゼにはアラニンラセマーゼのように補酵素としてピリドキサルリン酸を要求するものと、グルタミン酸ラセマーゼのように補酵素、補因子を全く要求しないものの2種類がある。α-アミノ酸のラセミ化という同じタイプの反応を触媒するこれら2種類の酵素の触媒機構がそれぞれどのように異なるのかを比較することは酵素学的見地から極めて意義深い。本研究は、両タイプの酵素の構造や機能を調べ、両者のそれぞれを比較することによってアミノ酸のラセミ化触媒の機構を総合的に解明することを目的として行われた。まず、ピリドキサルリン酸に依存するBacillus stearothermophilusの耐熱性アラニンラセマーゼについて、ピリドキサルリン酸を結合する39位のLysをAlaに置換した変異酵素を作製し、精製すると共に、その酵素化学的諸性質を調べた。特に、本変異酵素のピリドキサルリン酸との複合体の分光学的諸質性、β-クロロアラニンとの反応性を解析した。また、同じくピリドキサルリン酸依存性のPseudomonas putidaの低基質特異性アミノ酸ラセマーゼについて、遺伝子クローニングを行い、クローン株より本酵素を精製すると共に、酵素化学的諸性質、副反応であるアミノ基転移反応における水素転移の立体化学、触媒機構を解析した。一方、ピリドキサルリン酸非依存性のグルタミン酸ラセマーゼ及びアスパラギン酸ラセマーゼについては、部位特異的変異や各種SH試薬との反応性を調べ、グルタミン酸ラセマーゼの場合は、Cys73及びCys184のいずれが置換あるいは修飾されてもプロトン授受ができず、一方、アルパラギン酸ラセマーゼの場合には、2量体当りCys73が1残基修飾を受けると完全に失活することから、サブユニット2個で一つの活性中心を形成することを推論した。
