| Research Abstract |
(1)前年度、ウシ大動脈中膜ホスホリパーゼA(PLA)は、Ca^<2+>イオン存在下に4℃で日オーダのゆっくりしたタイムコースで活性化されることを見いだしたのでこの活性化に関与する因子の検索を行い、以下の結果を得た。a.活性化に対する2価イオンの効果は、Ca^<2+>>Sr^<2+>>Mg^<2+>であり、調べた限り他のイオンでは活性化は起こらなかった。b.プロ酵素の限定分解によるPLAの活性化の可能性を検討するためSHプロテアーゼ阻害剤、セリンプロテアーゼ阻害剤でPLAの活性化の効果を調べたところ、セリンプロテアーゼ阻害剤でPLAの活性化が著名に抑制された。c.グアニンヌクレオチド誘導体(GTP、GMP、cGMP、グアノシン)はPLAの活性化を促進した。d.NO発生剤であるNaN_3、NaNO_2、イソソルバイドはPLAの活性化を抑制した。
(2)PLAの精製収率の再現性が乏しく精製法の再検討をした。非イオン性界面活性剤C_<12>E_8の溶離液への添加により収率がある程度改善されることがわかった。
(3)大動脈PLAは、I群およびII群PLA_2と免疫学的に異なっていた。
