Research Abstract |
We have demonstrated a reciprocal translocation of t(2 ; 5)(p23 ; q35) in a case of Ki-1 positive human anaplastic large cell lymphoma. To investigate the breakpoint junction at molecular level, we have first performed PCR-based screening of YAC libraries by using three human genes of proopiomelanocortin (POMC, 2P23), apolipoprotein B (APOB, 2p23-2p24), and Nmyc (MYCN, 2p24.1) as screening probes. So far seven positive YAC clones (4 clones for POMC, 2 for APOB, and 1 for MYCN) were isolated from the Center d'Etude du Polymorphisme Humain (CEPH) YAC library. Analytical Pulsed-Field Gel Electrophoresis (PFGE) indicates that these YAC clones contain human inserts between 220 and 475kb. To test whether the YAC clones can detect the breakpoint or not, fluorescence in situ hybridization (FISH) analysis had been done by using Alu-PCR products as probes. POMC clone A7 (450kb) is suspected to be a closest clone to the breakpoint, didn't cover the breakpoint region and transferred to distal region of the derivative chromosome 5 (t5). Several cosmid clones mapped to 5q35(4 clones) and 2p23(16 clones) were also tested. One of these clones (cC12-496) is suspected to locate at the breakpoint region. Further studies are on going to verify the result of FISH.
