本年度の成果と検討
温熱療法による治療効果をより上げるため、癌細胞の細胞特性、特に癌関連遺伝子と温熱感受性とを結びつけた研究を進めてきた。特に癌抑制遺伝子p53の正常タイプおよび突然タイプの遺伝子産物は共に温熱処理によって細胞内量が増加すること、また細胞の温熱感受性に深く関わり温熱によって誘導されるヒートショックタンパク質72(HSP72)と結合することを明らかにした。また、cot、Ki-ras、Ha-ras、erbBの活性型癌遺伝子が癌細胞を温熱耐性にさせることを遺伝子導入法によって知った。さらに突然変異タイプのp53をもつ癌は放射線・温熱併用療法により、正常タイプp53をもつ癌より処理直後の治癒率が高いという予備的成果を得た。
今後の方向
このように癌関連遺伝子のタイプと癌細胞の温熱感受性を知ることは、温熱療法によるより効率のよい癌治療成績の向上につながると思われる。今後癌関連遺伝子産物がどのようなしくみで癌細胞の温熱感受性に影響するのか、そのしくみを明らかにする必要がある。
