Research Abstract	ヒト肝臓の糖タンパク質上のシアロ糖鎖にHB_S抗原粒子に対する受容体活性を見い出した。Q-SepharoseおよびSuperose6等のカラムクロマトグラフィーにより受容体活性を有する糖タンパク質を分離精製した。本糖タンパク質をSDS-ポリアクリルアミドゲル電気泳動後、PVDF膜に転写し、HB_S抗原粒子との結合性を確認した。さらに糖鎖特異性が既に明らかにされている種々のレクチンとインフルエンザウイルスを用いて糖鎖の解析を行った。その結果、分子量120-230KDの微量シアロ糖タンパク質にHB_S抗原粒子との結合性を確認した。また、インフルエンザウイルスの本糖タンパク質への結合特異性からHB_S抗原粒子との結合に、本糖タンパク質上のN-acetylneuraminic acid alpha2-6 Galactose構造が重要である可能性が示唆された。そこで本シアロ糖タンパク質画分を抗原としてBALB/cマウスに免疫し、モノクローナル抗体を作成した。本糖タンパク質に反応性が認められた抗体産生ハイブリドーマ中にHB_S抗原粒子との結合を阻害するものを認めたことから現在そのクローニングを行なっている。C型肝炎ウイルス受容体に関しては、ウイルス粒子が見いだされていないためバキュロウイルス系を用いてE,NS1/E2領域をカバーするタンパク質を発現している細胞を取得し、これを用いて新規受容体アッセイ系の確立を進めている。
