| Research Abstract |
本年度は、ニューロトロフィン-3(NT-3)の酵素免疫測定系を確立するために以下のことを行なった。
1.NT-3は抗原として多量に得ることができないだけでなく、種差がないためタンパクに対する抗体を得ることができない。そこで、NT-3のペプチド抗体作製を試みた。NT-3の多抗原性ペプチド(3カ所)とC末端のペプチドを合成し、ウサギに免疫し、大量の抗血清を得た。
2.NT-3特異抗体の精製に必要な抗原を、ヒトNT-3のcDNAを組み込んだCHO-N1/N2の培養上清から、S-セファロースおよびセファクリールS-100カラムによって精製した。
3.NT-3標品をカップリングさせたセファロースCL-6Bのアフィニティカラムを調整し、抗血清中のNT-3特異抗体の精製を用いて行なった。4つのペプチドのうち、3つに対して特異抗体が得られた。
4.得られた精製抗体を、常法にしたがって、beta-D-ガラクトシダーゼでラベルし、NT-3をスタンダードとして、酵素免疫測定を行なった。特異抗体が得られても、特にペプチド抗体の場合には酵素免疫測定系確立に有用でないことがしばしばある。現在のところ、C末端に対する抗体が酵素免疫測定法に使用可能であることが分かったが、測定系確立までにはいたっていない。
