| Research Abstract |
移植臓器特異的な寛容を導入する方法の一つとしてドナー由来の骨髄細胞による骨髄移植が有効であることが知られている。我々はドナー特異的な寛容の誘導の際のclonal deletionとclonal anergyの役割を明らかにすることを目的とし、限定された抗原とそれに対して反応するT細胞レセプターを認識するVbeta抗体を用いて骨髄キメリズム、T細胞のclonal deletion、そして皮膚移植片生着期間の関係を解析することを試みた。移植抗原としてはEalphaを、骨髄キメリズムのマーカーとしてはLy5のアロタイプを、またI-E反応性のT細胞の存在はVbeta11抗体により解析した。その結果、1)骨髄キメリズムの高さとVbeta11陽性T細胞のdeletionの程度には正の相関があること、2)皮膚片の長期生着には骨髄キメリズムが30%以上という閾値が存在することが明かになり、胸腺内でのT細胞のclonal deletionに必要な抗原量を定量的に解析する系を確立することができた。
