Research Abstract

細胞性粘菌の発生系を対象として以下の研究を行った。
形態形成の制御機構
細胞接着に関わる分子gp64とCsAの糖脂質および糖鎖の構造を明らかにし、細胞接着における役割を論じた(落合・吉田元信)。ブラストシジンS耐性株を用いる挿入突然変異体の分離法を確立し(田仲)、それを用いて有性生殖における細胞融合能の低い形質転換体を得た(柳澤)。細胞膜-細胞骨格標品を用いて、細胞運動関連蛋白質の動的構築変化を調べ、細胞内カルシュウムの関与を明らかにした(裕村恵彦)。cAMP刺激に対して細胞内カルシュウムが増加すること、予定柄細胞が予定胞子細胞よりも大きな反応を示すことを明らかにした(竹内)。細胞内cGMP量が細胞の高浸浸透処理によって増加することを見出した(小山正和)。
細胞分化の制御機構
飢餓処理時点での細胞の細胞周期上の位置が特異点(増殖・分化の分岐点)の直前にある細胞は集合中心として機能したのちに予定胞子細胞に分化し、直後の細胞は遅れて集合し、予定柄細胞に分化することを明らかにした(前田)。細胞の集合時に細胞密度に依存して発現する新たな遺伝子(dutA)を見出した。このRNAは翻訳されないが、広く他の生物にも存在する新しい種類のものである(岡本)。低細胞密度の細胞に分化を誘導する因子を培養上清中に見出し、それが70-80kDの蛋白質であることを示した。この因子は分化過程で細胞分裂をも誘導する(大畠明子)。予定胞子細胞に特異的に発現する遺伝子Dp87は集合前期の細胞にその位置と無関係に発現し、発現した細胞は移動体の予定胞子細胞域に選別される(田坂昌生)。
