Research Abstract	ブタ肺からインジュリン(HGF発現誘導因子)の精製を進めると、インジュリン活性を示す複数の分画に分離された。これら複数の分画の精製を進め、その性質を解析したところ、これらはPDGF、bFGF、IL-1であることを明らかにした。実際リコンビナントPDGFは線維芽細胞のHGFmRNAの発現を誘導し、HGFの産生を促進した。PDGFは血小板に豊富に存在しており、障害にともない血小板が活性化されると速やかに分泌される。したがって、組織傷害により血小板から分泌されるPDGFは線維芽細胞などの間質の細胞の増殖を促す一方、そのHGF産生を高め、HGFは主に上皮組織の再生を担うと考えられる。すなわち、PDGF→HGFはHGF/インジュリアンシステムの一つとして組織・器官の再生を担うネットワークの一つである。一方、bFGFに加え、aFGF,HST-1/FGF-4などFGFファミリーのいくつかはHGFの発現を誘導するHGFのup-regulatorであることを明らかにした。上記のようにHGFは上皮-間葉相互作用のメディエーターとして、肝臓、腎臓、肺などの器官形成、形態形成を担うことが明らかになってきたが、このような発生過程におけるHGFの発現誘導にはFGFファミリーに属する増殖因子が関与する可能性がある。その他、EGF、TGF-α、HB-EGF(heparin-binding EGFj-like growth factor)などのEGFレセプターのリガンド、プロスタグランジンも強力なHGF誘導因子であることを明らかにした。プロスタグランジンは傷害にもとない局所で産生され、炎症反応を誘起すchemical mediatorとして知られているが、おそらくプロスタグランジンは炎症反応を引き起こす一方でHGFの発現を誘導し、組織再生にも関与していると考えられる。
