細胞質に含まれる中性α-マンノシダーゼは糖蛋白質のエンド-β-N-アセチルグルコサミニダーゼの水解産物(糖鎖部分)を基質とし、糖鎖の代謝を行なっていると推測されている(奥、長谷 1992)。このマンノシダーゼは精製した時点では酵素活性が無く、コバルト(II)イオンとインキュベーションすることにより、酵素活性が増加する事が明らかとなった。上記の糖鎖代謝過程の役割を解明するためには、この酵素の基質特異性の解明とともに、コバルト(II)イオンによる活性化機構の解析が必要となる。このために量多く酵素を精製し、その物性や基質特異性等を調べ、さらにコバルト(II)イオンで活性化される機構の解明を試みる。
ウズラの酵素はこれまでに精製したものを使用する。しかし現有のウズラの酵素は本研究を行うには量的に不充分と思われるため、ニワトリ輸卵管よりコバルト(II)イオンで活性化される中性α-マンノシダーゼの精製を、ウズラの酵素の精製を参考にして行なう。ニワトリ輸卵管を超高速ホモジナイザー(キネマチカ社、Polytron)でホモジナイズし、高速冷却遠心機(J2-MC型、ベックマン社製)で遠心を行い上澄をとり、粗酵素標品とした。粗酵素標品より各種液体クロマトグラフィーを用いて酵素の精製を行ない、電気泳動的に単一な酵素とした。
