Research Abstract

Bovine oocytes matured in vitro was activated by various parthenogenetic stimuli (electrical or chemical). The higest activation rates were obtained when bovine oocytes were activated by electric pulse with electrolyte medium or by treatment with ethanol-cycloheximide.
Single non-viable spermatozoon was injected into ooplasm (ICSI) of bovine oocytes matured in vitro and cultured for 9 days in vitro. Karyotyping of resultant blastocysts was performed, and 8 of 9 possessed 60 chromosomes whereas 1 of 9 had 61 chromosomes. The frequency of blastocysts with an abnormal chromosome number (11%) was not higher than that observed in our standard IVF procedure (20%). Some embryos that developed to blastocysts were transferred to recipients and 4 calves (1 male and 4 females) were born. Two of these females have subsequently become pregnant, one by artificial insemination and the second via embryo transfer. Both females delivered male calves. This is the first demonstration of the normalcy of ICSI offspring in the post-puberal period, as evidenced by the birth of normal offspring from ICSI-derived females.
