| Research Abstract |
1.HLA-B51アロ反応性CTLクローンによるTAP依存性およびTAP非依存性アロペプチドの認識
T1,T2,.174および.174にTAP1とTAP2遺伝子を導入したトランスフェクタントを使い、9つのHLA-B51アロ反応性CTLクローンの認識するペプチドのTAP依存性を調べたところ、8クローンがTAP依存性で1クローンがTAP非依存性であることがわかった。
2.HLA-B51アロ反応性CTLクローンが認識するヒトのアロペプチドの分離
TAP依存性アロペプチドを認識するCTLクローンのうち1つのクローン(2B3)が認識するペプチドをHLA-B51分子から分離することができた。
3.ヒトマイナー組織適合(hmH)抗原特異的CTLクローンが認識するhmHペプチドの解析
HLA-B35分子に結合するhmHペプチドを以前にB細胞株より分離したが、さらに腎培養細胞から分離することができた。またHLA-B35分子の152番目をGluに置換したHLA-B35-152Eおよび171番目をHisに置換してHLA-B35-171H抗原はhmHペプチドを結合することができるが、hmH特異的T細胞によっては認識されないことを明らかにした。このことからHLA-B35分子のAポケットの構造変化がhmHペプチドの結合には大きな影響を与えないが、T細胞リセプターの認識に影響をあたえると考えられた。
