| Research Abstract |
B細胞抗原受容体はIgM,mb-1(Ig-α),B29(Ig-β)の3つの分子が細胞外に発現しmb-1,B29の細胞内部分に多くの機能分子群が結合していることが明らかになってきている。これまでにsrc type tyrosine kinase Fyn,Lyn,Blk,LckおよびSyk tyrosine kinaseが結合し抗原レセプターからのシグナル伝達をになっていることがあきらかになってきている。われわれは特にMB-1に結合する機能分子を検索し、それぞれのシグナル伝達経路がB細胞のどのような機能と関連しているのかを解析した。また、そのほかのどのようなシグナル伝達がこの抗原レセプターシグナル伝達に影響するのかを解析した。その結果まずsrc type tyrosine kinaseが活性化されその後Syk kinaseの活性化が誘導されるが、その際に関与する細胞内セカンドメッセンジャー分子が存在することを示唆する結果を得た。そして、それはB細胞の分化ステージによって異なることを示した(論文投稿準備中)。さらに続いて、われわれの見い出したp52分子も抗原受容体からのシグナル伝達に関与していることを示した(論文発表済)。この分子に対するモノクロナール抗体を用いてcDNAクローンを単離し、その遺伝子配列がこれまでに報告されていない新しい遺伝子であることを示した(論文印刷中)。この分子は顕著な機能ドメインを持たずSH3結合機能様モチーフを有することから新しいタイプのシグナル伝達機能分子の可能性が示された。
