| Research Abstract |
デオキシリボ核酸分解酵素I(DNase I)の遺伝的多型は、血清、尿および諸臓器(膵、肝、腎等)から判定可能であり、その遺伝子頻度のバラツキは大きく、個人識別能は極めて高い。DNase Iは、光、温度および湿度などに対しても極めて安定であり、厳しい条件下に暴露されている斑痕などの法医鑑定資料から実施する個人識別には極めて優秀なマーカーになるものと考えられる。最近、精液DNase Iにも遺伝的多型が存在していることが明らかにされた。さらに、DNase I活性は精液中には高いが、女性体液中に証明できないことが判明し、新たな精液の遺伝マーカーとしての応用が期待されている。今回の研究では、まず、ポリアクリルアミドゲル等電点電気泳動法とDNase I活性染色法のDAFO法とを組み合わせた精液斑からのDNase I型判定法を新たに開発した。また、精液斑のDNase I活性は0.3-1M尿素で効率よく抽出されることがわかった。次に、様々な条件下に放置して作成した多数の精液斑からDNase I型判定を実施したところ、-20℃、4℃および室温に約1年間放置してあった精液斑(精液量にして約0.3μlに相当)のいずれからも正確なDNase I型判定が可能であった。驚くべきことに、37℃下で湿箱中に13週間放置しておいた精液斑からでも正確な型判定が可能であった。さらに、実際の鑑定例の精液・女性体液混合斑痕についてDNase I型を検査したところ、いずれも精液の型のみが正確に判定できた。以上の実験結果から、DNase I型は法医鑑識分野の実際において有力な精液の個人識別マーカーとなり得ることが明らかになった。
