ヒト消化管腫瘍におけるガストリン受容体遺伝子の発現とその突然変異に関する研究

| Research Abstract |
1.ヒト・ガストリン受容体cDNAを用いた検討から、ヒト肺小細胞癌と肺小細胞癌株の約半数、大腸癌細胞株の10%、大腸癌の20%にガストリン受容体遺伝子の発現が認められた。一方、胃癌細胞株、胃癌には一例も発現がなかったが、胃小細胞癌株2例のうち1例(ECC10)に明らかな発現が認められた。
2.ガストリン受容体遺伝子の発現を認めたECC10細胞と大腸癌株LoVo細胞に対するガストリンの増殖効果を検討したところ、ECC10ではガストリンの増殖効果は認められなかったが、LoVo細胞はガストリンによって容量反応性の増殖反応を示した。
3.ヒト・ガストリン受容体遺伝子をN1H3T3細胞に導入したところ、本細胞はガストリンによって著明な増殖促進反応を示した。またこの際ガストリン受容体刺激は、MAPキナーゼ、focal adhesion kinase(P125^<FAK>)のチロシン・リン酸化を著明に促進させた。さらにこの際MAPキナーゼ活性の明らかな増加を認めた。
4.上記NIH3T3細胞では、ガストリン刺激によりc-fos、c-myc遺伝子発現の増加が認められた。
5.ガストリン刺激によりNIH3T3細胞のアクチン・ストレス・ファイバーの形成が促進され、細胞の可動性の増強が観察された。
