Research Abstract |
MULTIPLE SCLEROSIS (MS) IS CHARACTERIZED BY EXCERABATION AND REMISSION OF NEUROLOGICAL SIGNS CLINICALLY,AND FOCAL INFILTRATION OF LYMPHOCYTES AND MACROPHAGES IN THE WHITE MATTER,PATHOLOGICALLY.SEVERAL HYPOTHESES FOR ITS PATHOGENESIS INCLUDING VIRAL ORIGIN,AUTOIMMUE,GENETIC FACTORS AND OTHERS,HAS BEEN POSTULATED.AMONG THEM,WE INVESTIGATED T CELL RECEPTOR IN THE DEMYELINATING PROCESS TO FIND ANY CLUE IN THE PATHOGENESES OF MS.MACROPHAGE INFLAMMATORY PROTEIN (MIP) -1 alpha, MIP-1 beta AND RANTES ARE CLASSIFIED AS beta CHEMOKINES AND THEY ARE PRODUCED BY ACTIVATED MACROPHAGES IN TEH INFLAMMATORY AND DEMYELINATYING LESIONS.MIP-1 alpha CONTENTS IN THE CEREBROSPINAL FLUID (CSF) OF PATIENTS WITH MS AND CONTROLS WERE EVALUATED.USING ELISA KITS (R & D CO., U.S.A.). MIP-1 alpha CONTENTS IN THE CSF OF 18 PATIENTS WITH MS,13 IN ACUTE STAGE AND 5 IN CHRONIC STAGE,WERE 4.4 <plus-minus>5 PG/MIN., 1.9 <plus-minus>0.9 PG/MIN., RESPECTIVELY,AND SHOWED STATISTICALLY SIGNIFICANCE TO CONTROLS,8 PATIENTS OF NON-INFLAMMATORY NEUROLOGICAL DISEASES,0.3 <plus-minus>0.7 P IMMUNOHISTOCHEMICAL STUDY ON THE AUTOPSY BRAIN OF MS BY USING ANTI-MIP-1 alpha, MIP-1 beta AND RANTES WAS ALSO CARRIED OUT.IN THE ACUTE DEMYELINATING LESIONS, ASTROCYTES SHOWED POSITIVE ANTI-MIP-1 alpha STAINING.MACROPHAGES AND ASTROCYTES DISCLOSED POSITIVITY TO ANTI-MIP1 beta AND RANTES.THESE RESULTS ARE SUGGESTIVE THAT beta CHEMOKINES ARE PLAYING A IMPORTANT ROLES TO INDUCE THE INFILTRATION OF LYMPHOCYTES IN THE ACUTE DEMYELINATING LESIONS OF MS.
