Research Abstract	マウス骨髄細胞、脾細胞をIL-3を含む半固形培地中で培養すると種々のコロニーとともに多数の肥満細胞コロニーが形成された。1個の肥満細胞コロニーを釣り上げて、IL-3を含む液体培地に移し培養を続けることにより、比較的容易に単一細胞由来の大量の培養肥満細胞(BMMC)を得ることができるようになった。IL-3依存性のBMMCの増殖はIL-4,IL-10,stem cell factor(SCF)により相乗的に支持されることが明らかとなった。このようにして得られたBMMCはFcεRIの架橋により、脱顆粒するとともに、TNF,GM-CSF,IL-4を産生することが示された。ヒト肥満細胞の大量培養系は確立していなかったが、われわれはヒト臍帯血、骨髄より分離したCD34^+細胞をSCF存在下で液体培養することにより、ヒト肥満細胞の大量培養系を確立した。このCD34^+細胞からのSCF依存性の肥満細胞の産生は、初期を除きIL-3,GM-CSFにより著明に抑制され、IL-6,IL-11により促進された。臍帯血CD34+細胞をSCF、IL-6存在下で培養すると、増殖してくる細胞中に占める肥満細胞の比率は培養60日後にはほぼ80%に達し、培養100日以上経過するとほぼ100%となった。このようにして得られたヒモ培養肥満細胞は培養一年以上経過してもなお増殖力を保持している。最近、純化ヒト培養肥満細胞が大量に得られつつあるので、この細胞を用いてヒト肥満細胞もマウスと同様にサイトカインを産生できるかどうか検討を開始している。
