マウスおよびヒト肥満細胞からのサイトカイン産生機構に関する研究

Research Abstract	マウス骨髄細胞、腹腔細胞をIL-3+IL-4存在下に培養すると大量のマウス培養肥満細胞が得られた。ヒト臍帯血単核球から純化したCD34^+細胞をstem cell factor(SCF)とIL-6存在下で培養すると、培養2週間目よりtryptase陽性のヒト培養肥満細胞が出現し、培養を続けると次第にその頻度が増大した。培養50日目には約60%、100日目には99%に達した。肥満細胞増殖に有効なサイトカインの検討ではSCFは必須であり、IL-6あるいはIL-11点かによりその増殖が著明に促進された。一方、CD34^-細胞分画からは培養肥満細胞は得られないことから、肥満細胞前駆細胞はCD34^+細胞分画に存在することが明らかとなった。このようにして得られた培養肥満細胞は6か月以上の長期にわたって培養可能であり、細胞表面に発現しているFcεRIをIgEと特異抗原あるいはanti-IgEで架橋することによりヒスタミンを放出した。マウスおよびヒト培養肥満細胞を用いて、細胞表面のFcεRIをIgEと特異抗原あるいはanti-IgEにより架橋後、経時的にRNAを抽出し、種々のサイトカインのmRNAをreverse transcriptase polymerase chain reaction(RT PCR)あるいはNorthern blotting法により測定した。IL-3,IL-4,GM-CSFなどのmRNAが確認された。
