Research Abstract	本年度は以下の基礎的検討を行った。 1.、サイトカイン(特にIL2)cDNAをplasmid vector(pRc/CMV)を用いて、ヒト胃癌株化細胞に移入するtransfection assayを行うこと、およびその移入効率、安定性、DNA、蛋白レベルでの解析等行った。その結果、ある程度の効率で移入が認められサイトカン産生も認められたが、その期間は短期間(約60日)であった。したがって、より効率よく長期にわたる移入を行うには、viral vectorを用いることが必須と考えられた。 2.pBlueScriptに組み込まれたIL2cDNAを制限酵素で切りだし、LTRpromotorを持つレトロウイルスベクターpGEM5 neoに組換えた。この際、IL2cDNAの3'下流にあるpoly A signalを除くための工夫を行った。更に各種サイトカンcDNA(IL2,TNF-a,IFN-g)の組み換えを試みるとともに、現在packaging cellPA317へのtransfection assayを進めているところである。 3.無血清培地を用い、ヒト癌、特に各種消化器癌の初代培養を試み、4〜5種類の培養を得ている。更にSCIDマウスを併せて検討したところSCIDマウスを用いることによりより効率よく、培養株が得られる可能性が示唆された。この点についてさらに検索中である。
