| Research Abstract |
最終年度のまとめとして、以下の結果を得た。
1.サイトカイン(特にIL2)cDNAをplasmid vector(pRc/CMV)を用いて、ヒト胃癌株化細胞に移入するtransfection assayを行うこと、及びその移入効率、安定性、DNA、蛋白レベルでの解析等行った。その結果、ある程度の効率で移入が認められ、サイトカイン産生も認められたが、その期間は短期間(約60日)であった。従って、より効率よく長期にわたる移入を行うには、viral vectorを用いることが必須と考えられた。
2.pBlueScriptに組み込まれたIL2 cDNAを制限酵素で切り出し、LTR promotorを持つレトロウイルスベクターpGEM5 neoに組み換えた。この際、IL2 cDNAの3'下流にあるpoly Asignalを除くための工夫を行った。さらに、免疫接着因子B7-2のcDNAの組み換え体を作製した。次いでpackaging cell PA317あるいはφ2へのtransfection assayを行った。
3.無血清培地を用い、ヒト癌、特に各種消化器癌の初代培養を試み、4〜5種類の培養を得ている。更にSCIDマウスを併せて検討したところSCIDマウスを用いることにより、より効率よく培養株が得られる可能性が示唆された。
4.IL2あるいはB7-2を移入したpackaging cellと株化腫瘍細胞と混合培養し、IL2、B7-2産生腫瘍細胞株化を作製し、現在樹立しているところである。
