Research Abstract

Rapid immunofilter paper assay (RIPA) was applied for the diagnosis of the virus occurring in vegetatively propagative crops. RIPA was developed using latex beads coated with antibody to virus. White latex-coated antibody was immobilized as a solid in a line on the surface near the bottom of a filter paper strip (8x0.5cm). The bottom end of this paper strip was dipped for 1 min into a leaf extract from infected plant and then for 2 min into dyed latex coated with the virus antibody. A colored band appeared on the line where the white latex had been immobilized. RIPA is suitanlble for field diagnosis because that does not requie specialized equipment and is time-saving.
Konjak mosaic virus, dasheen mosaic virus and cucumber mosaic virus in konjak, chrysanthemum virus B in chrysanthemum, potato leaf roll virus, potato virus X and potato virus Y in potato, odontoglossum ringspot virus and cymbidium mosaic virus in orchids, garlic latent virus and onion yellow dwaft virus in welsh onion and strawberry pseudo mild yellow edge virus in strawberry were detected by RIPA.
Simple method of RIPA was developed by plotting antibody-coated white and dyed latexes on filter strip before testing. Another modification of RIPA to flow horizontally leaf extract and antibody-coated dyed latex was developed to detect of viruses from plants infected with multiple viruses.
