| Research Abstract |
トロンボキサン(TX)A_2とプロスタグランジン(PG)I_2はいずれもアラキドン酸から生合成され、血小板凝集や血管収縮にかかわる生理活性物質である。その生体内代謝動態を追究して病態生理学的意義を解明するため、代謝半減期の長い代謝物を測定するのがよいという考えから、11-デヒドロ-TXB_2,2,3-ジノル-TXB_2,2,3-ジノル-6-ケト-PGF_<1α>の免疫測定法の開発を試みた。
2,3-ジノル-TXB_2のα鎖のカルボキシル炭素をアルブミンに結合させたものを抗原とすると、TXB_2と高度に交叉反応する抗体が得られ、α鎖長の違いを認識しないものと考えられた。そこで2,3-ジノル-6-ケト-PGF_<1α>の場合は、ω鎖の末端をカルボキシル化したものを合成し、そこへウシ血清アルブミンを結合させたものを抗原として、マウスを免疫し、ハイブリドーマ法で抗2,3-ジノル-6-ケト-PGF_<1α>のモノクローン抗体を作った。幸にして特異性の高い抗体が得られ、6-ケト-PGF_<1α>との交叉反応率は0.01%以下であった。2,3-ジノル-6-ケト-PGF_<1α>は条件によってはγ-ラクトン型(閉環型)あるいはケトン型(開環型)として存在するが、検討の結果この抗体は開環型と反応することがわかった。この抗体を使って、2,3-〔^3H〕ジノル-6-ケト-20-カルボキシルメチル-PGF_<1α>を放射リガンドとして用いる放射免疫測定法の条件を設定した。また20-カルボキシル基をワサビのペルオキシダーゼに結合させた酵素標識抗原を作って、酵素免疫測定法の条件を設定した。
