| Research Abstract |
ヒトの上皮性のムチンの検出系と発現系を確立することを目的として、モノクロナルこうたいの作製、リコンビナント遺伝子を用いた発現ベクターの作製、及び培養細胞によるそれらの発現を行なった。(1)種類の異なるヒトムチンのコアペプチドの各種臓器の粘膜上皮に於ける発現量を検出定量できる抗体の開発を行うことを目的として、ヒトMUC1ムチンコアペプチドのC末端に近いグリコシレーションを受けない部分の15-20オリゴペプチドを合成し、これに対するポリクロナル抗体とモノクロナル抗体を作製中である。また、新しい抗MUC1ムチンモノクロナル抗体を作製した。(2)ムチンのコアペプチド(MUC1およびMUC2)のグリコシレーションを受ける部分を含むキメラ分子のリコンビナント遺伝子を作製した。これらをヒト上皮細胞に発現することに成功した。これらのグリコシレーションのパターンを解析中である。(3)一つの細胞集団由来のバリアント細胞で,MUC1ムチンの発現があるものとないものとを見いだし、発現がないものを用いてMUC1ムチン強制発現系を作製した。(5)ヒト腸内常在性細菌菌体から、大腸ムチンに結合する分子を単離しその一次構造を明かにした。これに基づき、遺伝子を取得,構造解析を行なっている。
