| Research Abstract |
以下のような操作を行うことのできる装置を試作した。
(1)倒立顕微鏡上で、生理的条件下の2個の細胞をマイクロペットで吸引補足できる。
(2)両者を一定時間接触させることにより、接着を成立させる。
(3)マイクロピペットの距離を広げることにより、細胞接着を引き剥そうとする。
(4)その時の細胞間にかかる力と接触面積の時間変化を測定・解析し、細胞間接着の特性を明らかにする。
試作された装置の詳細は以下の通りである。
(1)マイクロピペット間の変位の制御は、市販のマイクロマニピュレーターに圧電素子を組み込むことにより実現した。
(2)2細胞間の接着力の測定には、マイクロピペットのたわみを測定する方法と細胞を補足するための吸引圧力は圧力センサーでモニターした。
(3)細胞を保持するための吸引力を測定する方法の両者によった。
(4)細胞を一定速度または一定張力で引っ張れるようにした。
(5)顕微鏡はすべて光ディスクレコーダーに記録し、ビデオデジタイザーによる定量的解析を行った。
