Research Abstract |
Cytoskeleton mutants as well as the wild type of the basidiomycete Coprinus cinereus were analyzed, with the following results. 1.We examined, by electron microscopy, cells showing diffuse extension growth in the wild-type fruit body, and found cytoplasmic microtubules running mostly parallel to the cell axis, but did not find cortical microtubules, which are found in plant cells. 2.We examined the roles of astral microtubules in the movement and positioning of the nuclei and in septation during conjugate division, using a beta-tubulin mutant (BEN198), in which the formation of asters is inhibited. The results provided evidence against a direct involvement of astral microtubules in the positioning of the nuclei, and strongly suggested that astral microtubules control septation in conjugate division. 3.As part of a project to analyze beta-tubulin mutations at the levels of DNA and protein, we cloned and sequenced a wild-type beta-tubulin gene, which showed that the gene encodes 445 amino acids and has eight introns in it. 4.We discovered that actin is concentrated in hyphal regions where the nuclei sit in early phases of conjugate division, suggesting the involvement of actin in conjugate division. 5.Westernblotting analysis of mutants resistant to cytochalasin E (CE) strongly suggested that cer1, which can confer resistance to CE when mutated, encodes actin.
