| Research Abstract |
女性ホルモンであるエストロゲンは性分化や性周期、発ガンなどの生命現象に深く関わっているステロイドホルモンである。エストロゲン合成酵素(或はアロマターゼP450とも呼ばれる)はエストロゲンの生合成の律速酵素であり、生体内のエストロゲン量は本酵素の発現量によって制御されている。申請者は、ヒト絨毛性腫瘍細胞を用いて、代表的な発がんプロモーターである12-O-tetradecanoyl-phorbol 13-acetate(TPA)による本酵素遺伝子の転写の誘導機構を分子レベルで解明する目的で本研究を行った。この転写誘導には本酵素遺伝子の転写開始点の上流2141-2098の位置にある44塩基対の配列が必須であることが明らかになっていた。そこで、転写誘導に関与する核内因子が結合するために必要な最小限のmotifを明らかにするたに、44塩基対の種々の領域をカバーするoligonucleotideを合成し、gel retardation assayおよびCAT assayをった。その結果、TPAによる転写誘導には-2141と-2115の領域にある26塩基対が必要であること、そしてこの26塩基対には今までに報告された転写の制御配列とhomologyを示す配列がないことが判明した。さらに細胞へのTPA処理の時間に依存して、この26塩基対に結合する核内因子が高分子化し、この高分子化と本酵素遺伝子の転写誘導との間に相関関係があることが明かとなった。申請者は今後、この核内因子の特性をさらに究明するため、因子の精製さらにはそのcDNAの単離を行う計画である
