Research Abstract

The human genome project and cDNA project are providing enormous amount of nucleic acid sequences and predicting genes and proteins. These proteins should be demonstrated to be real gene products and characterizad with their forms in the cells. We are trying to isolate proteins in the human cells by high resolution two dimensional electrophoresis and provide their amino acid sequences. Our two dimensional electrophoresis with immobilized pH gradient strip for isoelectric focusing and with large format gel (22cm x 22cm) for second dimension gives about 2,000 to 2,800 spots for whole cell extracts. Two dimensionsl gel electrophoresis pattern were obtained from the following various human originated cell lines ; HeLa S3, HeLa P3, Epidermoid carcinoma (KB), Ovarian carcinoma (BG-1), Hepato cellular carcinome (HepG2), Hepatoma PLC/PRF/5, Promonocytic cell line (u937), Gastric cancer (HGC). Lung adenocarcinoma (HLC), T cell line (HPB-ALL,JurKat, PEER,MOLT-4), B cell line (NALM-6, BALL-1), Myelomonocyte cell line (ML-2, HL60), Retinoblastoma (Y79). some of nuclear proteins and some proteins from the whole cell from HeLa were determined by micro sequencing.
