胃癌の進展・転移に深く関与する6.0Kbc-met異常転写産物の分子クローニング

Research Abstract

1.平成7年度の研究経過
(1)胃癌ならびに正常胃粘膜におけるC-met遺伝子スプライス変異体の解析を行い、これら組織においてはc-met cDNAのnt2265からnt2319までの小エクソンがスプライスアウトされた形のトランスクリプトが主として発現されていることを明らかにし、論文発表した(Yokozaki, H., et al. Oncology Reports, 3, 1996(in press))。
(2)胃癌細胞株cDNAライブラリーならびにpAP3neoライブラリーのスクリーニングを前年度に引き続き継続したが、c-met6.0Kbトランスクリプトはそのサイズが大きいこととさらに稀少トランスクリプトであること等より、陽性クローンを得ることは出来なかった。
(3)前年度より、ヒト抹消血白血球genomic libraryからのクローニングを試みたが、(2)と同様に陽性クローンを得ることは出来なかった。
2.平成7年度の研究の評価
以上の如く、通常の方法によってはc-met6.0Kb異常転写産物の分子クローニングは成功せず、long PCR等の高感度のRT-PCR法による直接クローニングの検討が必要なものと考えられた。
