Research Abstract

ハプテン(trinitrophenol=TNP)特異的間質性腎炎の誘導実験において、単量体の精製陽性荷電化TNP-ovalbumin(PI>10)を、ラット並びにマウスの左腎動脈から選択的に腎灌流を行なった。細胞性免疫負荷実験では、抗原投与の7日前に4%塩化ピクリルエタノール溶液を、ラット腹壁皮内注入ないしはマウス腹壁に塗布した。抗原投与の24時間後、尿細管上皮に顕著な壊死性変化を認めたが、間質内炎性細胞浸潤はなく人体症例に見られる尿細管炎の病像は得られていない。負荷のない対照群では、TNPによる非特異的細胞傷害性病変を認めた。液性免疫負荷実験においては、抗原投与の24時間後、抗TNP-ヒトIgG家兎血清を経静脈的に投与した。24時間後、好中球を主体とした尿細管間質炎を認め、In situ型間質性腎炎が発症した。細胞移入実験では、TNP感作マウス脾細胞から、全T細胞、CD4陽性細胞、CD8陽性細胞を分離し、そのそれぞれを、抗原投与1日後のマウスに細胞移入したが、36時間後の腎には、尿細管間質炎は認められなかった。
