| Research Abstract |
小児伝染性紅斑の原因ウイルスとして知られているヒト・パルボウイルス(HPV/B19)は、近年、非免疫性胎児水腫や慢性関節リウマチなどの他の重篤な疾患との因果関係が指摘されている。しかし、HPV/B19感染の血清診断法の確立は充分ではなかった。我々は今までに、HPV/B19ウイルス粒子を抗原とするELISA法、抗HPV/B19モノクローナル抗体を用いたウエスタン法、PCR法を用いたHPV/B19 DNAの検出法を確立してきた。
本研究では、まず大腸菌で産生したグルタチオンSトランスフェラーゼ(GST)と融合したHPV/B19 VP-1蛋白を抗原とした、抗HPV/B19抗体のELISA法を用いて抗HPV/B19血清の測定値と、同じ抗原を用いたWestern blot法の結果を比較した。ELISAのOD値が0.1以下検体では、OD値とWestern blot法による陽性とはかならずしも相関しなかった。この境界域を明確にするために、ELISAのOD値0.1から0.01までの73検体について詳細に検討した。その結果、OD値が0.08以上のものはWestern blot法ですべて陽性であった。0.04-0.08の検体の一部はWestern blot法で陽性と認められた。従って、本ELISA法でOD0.08以上は抗HPV/B19陽性と判定できるが、境界域のOD値0.04-0.08を示す検体はWestern blot法での確認が必要である。
