Research Abstract	初乳中の胃粘膜増殖分化因子の精製をおこなうために、胃表層上皮粘液分泌細胞に分化しうる胃未分化上皮細胞の初代培養系を確立した。ラット胃粘膜より単離した胃粘膜細胞をcounterflow elutriationを用いて分離し、強いc-fos発現とDNA合成のみられる未分化増殖上皮細胞を得た。本細胞を培養すると3日後には、confluentなmonolayerとなり、わずかに粘液を産生したが、胃粘膜の他の上皮細胞への分化を示す遺伝子発現はみとめられなかった。このconfluent monolayerに、初乳の乳清を10%含むmediumを添加すると上皮細胞の急速なDNA合成の増加と粘液産生細胞への分化がみとめられた。さらに、粘液分泌の増加と一致してPGE_2のEP_2受容体の遺伝子発現の亢進がおこることが明らかとなった。そこで、胃粘膜上皮細胞のDNA合成促進、粘液分泌亢進、PGE_2、EP_2受容体遺伝子発現を指標に、乳清に含まれる増殖、分化因子の精製をこころみた。まず、乳清を抗ラットEGF抗体を結合させたアフィニティーカラムに通し、EGFを除いた後に、Mono-Qイオン交換カラム、ヘパリンセファロースアフィニティーカラム、ゲルロ過カラムを用いて分画をおこなった。胃粘膜上皮のDNA合成を促進する物質は、ヘパリン結合性を有する分子量約3万の蛋白質であり、HGF、FGF、KGFとは異なった未知の物質であった。一方、粘液分泌亢進とPGE_2受容体の発現を亢進させる物質は、分離することができず同一の物質であると考えられた。この物質はヘパリン結合性のない分子量約2万の蛋白質で糖鎮を有すると考えられた。現在、この2種の蛋白質のアミノ酸分析をすすめており、ひきつづいてcDNAクローニングをおこなう。
