| Research Abstract |
Alkaline phosphataseは細胞膜輸送に関係した酵素と考えられている。ヒト汗腺におけるalkaline phosphataseの分布を酵素組織化学的に明らかにした。Alkaline phosphatase活性はエックリン汗腺では細胞間微小汗管に限局して存在していた。この結果は細胞間微小汗管は単に腺腔側細胞膜の表面積を増大させるだけではなく、機能的に分化した構造であり汗産生に深く係わっていることを示している。アポクリン汗腺では筋上皮細胞膜の分泌細胞に接している部分と、分泌細胞膜の基底膜側で筋上皮細胞に接している部分に酵素活性が認められた。エックリン汗腺とアポクリン汗腺の筋上皮細胞ではalkaline phosphatase活性の分布が異なっており、これは両汗腺の機能的差を反映したものと考えられた(J Histochem Cytochem 43:927-932,1995)。
エックリン汗腺あるいはアポクリン汗腺の分泌部を構成している細胞が増殖しているか否かを検討した。さらに、分泌部を構成しているどの細胞が増殖しているかを明らかにした。増殖細胞の検出は、単離した汗腺組織をin vitroで3気圧95%O_2-5%CO_2中でのbromodeoxyuridine標識(Morimoto & Saga:Br J Dermatol 125:9-13,1991)と、PCNAの免疫染色により行った。エックリン汗腺とアポクリン汗腺のいずれかにおいても分泌細胞は増殖していたが、筋上皮細胞は増殖していなかった。これらの結果は分泌細胞は一定の割合で失われ、分泌細胞自身の増殖により補われていることを示している(J Histochem Cytochem 43:1217-1221,1995)。
