Research Abstract |
IN 1993, THE TRIAL TO APPLY MONOCLONAL ANTIBODY BOUND ANTISENSE DNA FOR SELECTIVE IMMUNOTARGETING GENE THERAPY OF COLON CANCER HAD BEEN CONDUCTED, THOUGH THE RESULTS WERE NOT SATISFACTORY.IN 1994, ANTISENSE DNA INTORDUCED INTO RETRO-VIRUS THOSE WERE COATED WITH MONOCLONAL ANITOBODY AGAINST COLON CANCER HAD BEEN DEVELOPED.THE HUMAN COLON CANCER CELL LINES WERE INFECTED AND THE EXPRESSION OF c-myc GENE WAS EXAMINED.THE EXPRESSION OF c-myc GENE WAS INHIBITED IN ANTIGEN POSITIVE CANCER CELL LINES BUT NOT IN CELL LINES WITHOUT ANTIGEN.THE SIZE OF MONOCLONAL ANTIBODY CHANGED THE AFFINITY OF ANTIBODEY RESULTED IN DECREASED EXPRESSION OF ANTISENSE DNA IN TARGET CELLS.TEH MOST IMPORTANT PROBRLEM TO OVERCOME SEEMED POOR EFFEICENCY OF VIRUS INFECTION.
