ヒト悪性神経膠腫におけるサイトカイン・成長因子遺伝子発現異常に関する研究

Research Abstract

本研究により,多くの成果が得られている.
悪性神経膠腫(以下glioma)細胞がinterleukin‐8(IL‐8)とくに77アミノ酸型のIL‐8を産生することを見い出し,誌上発表を行っている(Tada M・他J Neuro‐Oncol 15:25).
また,tumor necrosis factor‐α刺激によりglioma細胞がIL‐6,IL‐8,Mn‐superoxide dismutase,IL‐1などの種々の因子を産生することも見い出し,誌上発表を行った(Sawamura Y・他J Neuro‐Oncol 15:197).さらにglioma細胞はgranulocyte‐macrophage colonystimutating factor(Sawamura Y・他Neurol Med Chir 33:603),monocyte chemoattractant protein-1(Tada M・他Int J Cancer 58:240),interleukin-1 receptor antagonist(Tada M・他J Neuroimmunol 50:187)を産生することを見い出しそれぞれ,誌上発表を行っている.こうしたglioma細胞が産生するサイトカインについては総説として論じている(Tada M・他J Neuro‐Oncol 17:261).また,これらのサイトカイン・成長因子が実際のglioma患者の髄液中に検出され,診断に有用なことも見い出し報告している(沢村豊・他神経免疫研究8:204).受容体に関しては,RT‐PCR法を用い,glioma細胞がIL‐1 receptor type l,type2;p55,p75 TNF receptor;interferon α/β,γ receptor,GM‐CSF receptor,MCSF receptor,SCF receptor,IL‐8 receptorを発現することを見い出し誌上発表している(Tada M・他J Neurosurg 80:1063;Sawamura Y・他Neurol Med Chir 35:567)
