Research Abstract

The purpose of this project have been to make pre-cancerous region and carcinogenesis, which is seen high frequency in patients of oral and maxillofacial field, by chemical reagent in animal models. We intended to analyze original and metastatic region immunohistochemically, but we failed to make carcinoma in rat models, by the method of using DMBA reagent which was performed in hamster model. Therefore spontaneous and transplantable rat squamous cell carcinoma, FF-6, was used to examine the tumor metastatic mechanism, change of regional lymph node weight, and histological change using a newly made monoclonal antibody, UB-17. Whether UB-17 antigen was recognized or not in the serum and ascites of carcinoma bearing rats.
The results were below. (1) UB-17 antibody reacted not only to FF-6 rat squamous cell carcinoma but also to human squamous cell carcinoma, and a part of normal rat and human squamous epithelium. This antibody may react to non-tumor specific antigen. (2) The characterization of this antigen was now unclear because of unstablity against many chemicals and heat, and molecular weight of this antigen was not determined. Ultrastructural localization of UB-17 antigen was on the cell surface of horny cell layr of squamous epithelium examined using the equipment of ultra-violet polymerization. (3) After transplantation of small piece of FF-6 to the lip and tongue of rats, the weight of regional (superficial) lymph node gradually increased. Low differentiated squamous cell carcinoma was seen in metastatic lymph node at 2 weeks after transplantation into tongue. Then it changed the histological structure, to well differentiated type at 4 weeks later. (4) UB-17 antigen was recognized in the serum and ascites of FF-6 bearing rats examined by radioactive binding inhibition assay. The result suggested that UB-17 antibody may be useful for serological diagnosis in patients instead of histological biopsy.
