Research Abstract

1, We investigated the effects of interferon-gamma (IFN-gamma) and tumor necrosis factor-alpha (TNF-alpha) treatments of NR-S1 cells (morine squamouse cell carcinoma). Since NR-S1 cells treated by combination of IFN-gamma and TNF-alpha were increased susceptibility to conjugation and lysis by lymphokine activated killer (LAK) cells, we examined the participation of cell-adheision molecules on the NR-S1 cells. Untreated NR-S1 cells expressed MHC class I,MHC class II,intercellular-adheision molecule-1 (ICAM-1) and ICAM-2. Treatment with IFN-gamma induced the expression of MHC class I and ICAM-1 molecules, but did not affect the expression of MHC class II and ICAM-2 molecules on NR-S1 cells. Treatment with TNF-alpha induced the expression of ICAM-1 molecule, but did not affect the other molecule's expression. However, treatment with the combination of IFN-gamma and TNF-alpha induced strongly expression of MHC class I and ICAM-1 molecules, slightly expression of ICAM-2 molecule compared with IFN-gamma or TNF-alpha alone. As the results of the expression of these molecules, NR-S1 cells became to be more adheisive to LAK cells and more susceptibility lysis by LAK cells.
2, Three patients with oral cancer were treated by peritumoral injection of IL-2 and IFN-beta every day for ten days. Finised injections, enhancement of NK actitity of patient's peripheral blood lymphocytes occurred, the percentage of IL-2 receptor, CD3, CD8 and CD56 positive cells in peripheral blood lymphocytes increased.
